Flow cytometric analysis of Anti-BCMA (C11D5.3) CAR 293 cells staining with Monoclonal Anti-Whitlow 218 linker Antibody, Rabbit IgG (P3C09), CF (carrier free), premium grade (Cat. No. 218-MY23G0) at 1.6 μg/mL (0.16 μg of the antibody stock solution corresponds to labeling of 1e6 cells in a final volume of 100 µL) and Goat anti-Rabbit IgG (H+L), PE. PE signal was used to evaluate the binding activity (QC tested).

No. 218-MY23G0) binding to CD3+ cells present in human PBMC. 5e5 of human PBMCs were simultaneously stained with FITC anti-CD3 antibody and Monoclonal Anti-Whitlow 218 linker Antibody, Rabbit IgG (P3C09), CF (carrier free), premium grade (0.16 μg of the antibody stock solution corresponds to labeling of 5e5 cells in a final volume of 100 µL) and washed and then analyzed with FACS. Both FITC and PE positive signals was used to evaluate the non-specific binding activity to human CD3+ cells (QC tested).

  • Background
    The whitlow/218 linker, with the sequence GSTSGSGKPGSGEGSTKG, is a commonly used synthetic peptide linker. Whitlow/218 linker often leveraged to connect the variable heavy (VH) domain and variable light (VL) domain of single-chain variable fragments (scFvs) and chimeric antigen receptors (CARs) that utilize an extracellular domain scFv for target antigen recognition. The scFv containing the 218 linker showed reduced aggregation and was found to be more stable to proteolysis in vitro.
